Interrelation of actions of glucose, insulin, and glucagon on induction of adenosine triphosphate: D-hexose phosphotransferase in rat liver.

چکیده
Insulin does not modify the low levels of liver adenosine triphosphate : hexose phosphotransferase attained in rats fed a carbohydrate-free diet, nor does insulin accelerate the induction of phosphotransferase by glucose, indicating that endogenous hormone is sufficient. Glucagon prevents the induction of ATP: hexose phosphotransferase, and this effect is not counteracted by an excess of insulin. A small dose of glucagon that has only a slight effect on phosphotransferase induction causes a definite glycogenolysis in the liver. Furthermore, certain observations on the levels of liver glycogen in animals receiving insulin indicate that secretion of glucagon secondary to insulin hypoglycemia does not interfere seriously with enzyme induction. A hypothesis is presented in an attempt to understand the interplay of different factors affecting ATP: hexose phosphotransferase activity in liver. Glucose and glucagon are considered as inducer and repressor, respectively. Insulin might be a permissive agent, probably acting at the inner membranes of liver cells.

THE EFFECTS OF GLUCAGON, INSULIN AND S TEROID HORMONES ON PHOSPHATIDATE PHOSPHOHYDROLASE ACTIVITY IN RAT LIVERS
The effects of steroid hormones, glucagon and insulin on rat liver phosphatidate phosphohydrolase (PAP) activity were studied both in vitro and in vivo. Incubation of rat hepatocytes with each hormone showed that dehydroepiandrosterone (DHEA), progesterone and testosterone increase PAP activity by 44.6, 37 and 36.9%, respectively. Estradiol, however, decreased enzyme activity by 13.6% under...

Peptides derived from the glucagon gene Gcg, for example, glucagon and glucagon-like peptide 1 (GLP-1), act as physiological regulators of fuel metabolism and are thus of major interest in the pathogenesis of diseases, such as type-2 diabetes and obesity, and their therapeutic management. Glicentin-related pancreatic polypeptide (GRPP) is a further, 30 amino acid Gcg-derived peptide identified ...
متن کاملTHE EFFECT OF SULFUR MUSTARD ON GLUCOSE PHOSPHORYLATING ENZYMES AND LIVER CELLS IN MALE RATS
A significant decrease of hepatic high-Km glucose- phosphorylating enzyme glucokinase (ATP- D- glucose 6- phosphotransferase, EC 2.7. 1.2, GK) and low-Km hexokinases (EC 2.7. 1.1. HK) activities were noticed in liver treated with 2, 2'- dichlorodiethyl sulfide (sulfur mustard, HD) when injectedinteraperitoneally to male rats in 114 LD50. This decrease occurred 1.5 to 5.5 hours after the in...
متن کاملModulation of Hepatic Gluconeogenesis: A Therapeutic Target for Diabetes
Liver is the major organ responsible for the maintenance of glucose homeostasis under fasting conditions. Hepatic gluconeogenesis is tightly regulated by the complementary actions of insulin and glucagon, the principal hormones that respond to feeding status. In the fasted state, glucagon secreted from pancreatic alpha cells promotes the synthesis of cyclic adenosine monophosphate (cAMP) from a...
متن کاملThe amino acid sequence of rat liver glucokinase deduced from cloned cDNA.
Rat liver glucokinase (ATP:D-hexose 6-phosphotransferase, EC 2.7.1.1) was purified to homogeneity, cleaved, and subjected to amino acid sequence analysis. Forty-five percent of the protein sequence was obtained, and this information was used to design oligonucleotide probes to screen a rat liver cDNA library. A 1601-base pair cDNA (GK1) contained an open reading frame that encoded the amino aci...
